Platelet signalling pathways
|
| I am focusing on elucidating signalling pathways and key enzymes implicated in activation and aggregation of human platelets. Activation of platelets by a primary agonist, such as thrombin, involves the secretion of pro-aggregatory mediators, including the adenine nucleotides ADP and ATP, as well as the generation and release of lipid mediators such as TXA2. Once released, these mediators initiate stimulatory loops by activating their respective platelet receptors, thereby representing important reinforcement mechanisms for platelet functions.
We are interested if and how thrombin receptor subtypes, PAR1 and PAR4, differentially induce platelet activation, secretion, and aggregation, and to what extent these responses are dependent on subsequent signalling pathways induced by the secondary mediators ADP, ATP, and TXA2.
